Discovery of Anastrozole
Anastrozole, a drug originally developed by Imperial Chemical Industries (ICI), was patented in 1987 and approved for medical use in 1995 for breast cancer treatment. Anastrozole is the type of hormone for hormone therapy and is sold under the brand name Arimidex. This drug is used for the best treatment of cancer as breast cancer and women going through menopause and post-menopause symptoms. Anastrozole belongs to the class of medications as non-steroidal aromatase inhibitors. Anastrozole works by decreasing the total amount of estrogen which the body produces.Â
Mechanism of Action
As discussed earlier, anastrozole is the non-steroidal AI drug that inhibits the aromatase enzyme’s functioning when it binds to the heme ions to reverse the function. By inhibiting aromatase activity, this drug helps reduce the levels of E1, E2, and E1S in the mammary tissues and the periphery. AIs (aromatase inhibitors) tend to lower estrogen levels by inhibiting the aromatase enzyme in the fat tissues. Aromatase changes the different body hormones into estrogen. In the case of breast cancer, estrogen can fuel the intensified growth of different breast cancer cells. Anastrozole doesn’t inhibit the production of estrogen from the ovaries; rather, it only prevents the conversion of different hormones to estrogen. Anastrozole is the hormone that is used for the alternative of different treatments for hormone replacement therapies in men and women. High estrogen and estradiol levels among women and men can cause different disorders and dysfunctions.
Clinical Applications:
The most common indications of anastrozole include:
Breast cancer:
Anastrozole is the first line of treatment for hormone-receptor-positive breast cancer in its early stages. It also has a first-line indication of unknown and hormone receptor-positive metastatic disorders or breast cancers. Anastrozole also has a potent indication for the treatment of advanced breast cancer, which has become resistant o the treatment with tamoxifen. Moreover, anastrozole has only a good prognosis or strong use for women in their post-menopausal phase.
Generally, most breast cancers and tumors grow in relative response to estrogen. Anastrozole helps in interfering with estrogen synthesis or production in the body. As a result of this phenomenon, the estrogen amount exposed to the tumors is greatly reduced to limit the tumor growth in the body.
Treatment of male infertility:
For male fertility, anastrozole is the most commonly used drug in men suffering from elevated estradiol levels (as more than 60 pg/mL) or even the abnormal ratios of testosterone and estradiol (less than 10:1). For such men, anastrozole is an effective drug in improving abnormal semen parameters. According to scientific studies, even a mild increase in estradiol concentration plays a relatively negative role in men’s health, resulting in sexual dysfunction and libido.
