Discovery:
Noopept or GVS-111 is a member of the racetam family. However, it is not considered to be a true racetam. This compound was developed in Russia, where it is commonly called Ноопепт. A Russian-based pharmaceutical company called JSC LEKKO synthesized Noopept in 1996. It was developed from the endogenous neuropeptide cycloprolyglycine. The effects of Noopept were very similar to Piracetam. However, it is 1000 times more potent than Piracetam.
Mechanism of action:
The exact mechanism of action of Noopept is still unclear. However, several mechanisms have been suggested for Noopept.
- Noopept metabolizes into cycloproglycine which has nootropic potential.
- Administration of Noopept results in an increase in nerve growth factor. This is essential for the growth and survival of neurons. Moreover, it increases the brain-derived neurotrophic factor that helps in the growth of new neurons and synapses.
- The anti-anxiety properties of Noopept come from the increase in tonic inhibition in the hippocampus.
Clinical Applications of Noopept:
Alzheimer’s Disease
According to research, a 7-day treatment of noopept improves fear memory. Moreover, it also prevents the amnestic effects of cholinergic neurotransmission inhibitors (Scopolamine and nicotinic receptors antagonist). This research suggests that this may be one of the mechanisms to boost cholinergic transmission. Noopept also produces a dose-dependent anti-amnestic effect.
Anxiety
According to research, Noopept helps in reducing symptoms of anxiety and improving the duration of sleep. This effect is evident from the first week of treatment, patients reported a reduction in increased irritability. From the second week of treatment, patients had less daytime drowsiness. In the third week, the patients had less anxiety and a depressed mood. Moreover, there was also a decrease in headaches. This is evident that noopept has a wide spectrum of anxiolytic and psychostimulatory effects.
Alcohol-induced cognitive disturbances
According to research, Noopept helps in the prevention of impairment of working memory that is due to the withdrawal of ethanol. Noopept helps in the restoration of glutamate and GABA in the hippocampus. This effect is different from its parent drug, piracetam. Because of these effects, Noopept is useful in treating alcohol-related behavioral disorders.
